  • Steps solidifying a role for SEPT9 in breast cancer suggest that greater strides are needed

    Human septins are a conserved family of GTPase proteins, encoded at thirteen loci scattered throughout the human genome. Of these, SEPT9 has been most widely implicated in diverse cancers of epithelial origin, including breast cancer, as well as leukemia. In this issue, Connolly et al. present compelling data that further supports a role for involvement of SEPT9 isoforms early in mammary tumorigenesis as well as intriguing evidence that suggests that different cellular localization patterns of SEPT9 isoforms may be a contributing factor in oncogenesis.

  • NF-kappa B, stem cells and breast cancer: the links get stronger

    Self-renewing breast cancer stem cells are key actors for perpetuating tumour existence and for treatment resistance and relapse. The molecular pathways required for their maintenance start to be elucidated. Among them is the transcription factor NF-kappa B, which is known to play critical roles in cell survival, inflammation and immunity. Recent studies indicated that mammary epithelial NF-kappa B regulates the self-renewal of breast cancer stem cells in a model of Her2-dependent tumorigenesis. We will describe here the NF-kappa B-activating pathways that are involved in this process and in which progenitor cells this transcription factor is actually activated.

  • Second malignancies in breast cancer patients following radiotherapy

    A study, evaluating the effect that radiotherapy may have had on the subsequent risk of second malignancies, including the possible influences of age at treatment and menopausal status, indicates that a raised risk of second malignancies is associated with radiotherapy for breast cancer, particularly for women treated after the menopause.
